Abaloparatide

Overview

Abaloparatide (Tymlos) is a synthetic analog of parathyroid hormone-related protein (PTHrP) FDA-approved for osteoporosis treatment in postmenopausal women at high fracture risk.

Mechanism of Action

• PTH1R agonism — Selectively activates parathyroid hormone receptor 1
• Bone formation — Stimulates osteoblast activity and new bone formation
• Cortical bone — Preferentially builds cortical bone over trabecular
• Fracture reduction — Reduces vertebral and nonvertebral fractures

Dosing

80mcg subcutaneously once daily. Max 2 years of treatment per FDA label.

Safety

FDA-approved. Risk of osteosarcoma (boxed warning). Contraindicated with Paget's disease, prior radiation, or bone metastases.
